CHAPS是一种非变性两性离子去垢剂,常用现在细胞裂解液中,可促进膜蛋白的溶解。其紫外吸收低,便于用紫外吸收法定量蛋白,是常用去垢剂。其分子量仅为615,经脱盐、超滤或透析可方便地除去。常用于等电聚焦和双向电泳等实验。


CHAPS is a non-denaturing, zwitterionic sulfobetaine detergent for solubilizing membrane proteins and

breaking protein-protein interactions.Commonly used for protein solubilization in isoelectric focusing and two-dimensional electrophoresis,3-5 especially for nondenaturing (without urea) IEF. CHAPS has been shown to give excellent resolution of some subcellular preparations and plant proteins. Concentrations between 1-4% (w/v) are typically used in an IEF gel.

A commonly used IEF sample solution consists of 8 M urea, 4% CHAPS, 50-100 mM dithiothreitol (DTT), and 40 mM Tris.9 Furthermore, its small micellar molecular weight (6,150) and high critical micellar concentration (6 - 10 mM) allow it to be removed from samples by dialysis.

As a zwitterionic surfactant,CHPS is used in the laboratory to solubilize biological macromolecules such as proteins. It may be synthesized from cholic acid and is zwitterionic due to its quaternary ammonium and sulfonate groups; it is structurally similar tocertain bile acids, such astaurodeoxycholic acid and  taurochenodeoxycholic acid. It is used as a non-denaturing  solvent in the process of protein purification and is especially useful in purifying membrane proteins, which are often sparingly soluble or insoluble in aqueous solution due to their native hydrophobicity.

CHAPS is an abbreviation for 3-[(3-cholamidopropyl)dimethylammonio]-1- propanesulfonate. A related detergent, called CHAPSO, has the same basic chemical structure with an additional hydroxyl functional group; its full chemical name is 3-[(3-cholamidopropyl)dimethylammonio]-2-hydroxy-1-propanesulfonate. Both detergents have low light absorbance in the ultraviolet region of the electromagnetic spectrum, which is useful for laboratory workers monitoring ongoing chemical reactions or protein-protein binding with UV/Vis spectroscopy.

Application

CHAPS is a zwitterionic surfactant for solubilization of membrane proteins. Combines the properties of both the sulfobetaine-type and bile-acid detergents.
